This is usually what I refer too for sizes.

However I must admit that with my most recent sprites I'm ignoring those sizes for gameplay reasons.

Koopaul wins. And Chaoxys, you have a good point: it's all about gameplay use.

K, just to make things clear, the following sprite isn't meant to be the in game sprite, I mainly made it to get a general design down. Basically, I want some opinions on whether or not it looks fitting for a Mario game.
Of course I won't say no to any general C+C as it follows the general rules of the style, meaning I could work with it to make it the in game sprite, but my main concern is knowing if it looks fitting.
Mega Drill, a successor of sorts to SMG2's Mega Hammer. It's the boss of world 9 and is fought while riding minecarts. It starts in the vehicle form and later goes to the standing form.

A number of them actually. The boss of World 8 is a reference to DK, one vertical level has numerous DK (arcade) styled layouts, barrels included, and then you have the mine carts.

Bowser Jr. in the Mini Koopa Clown hovercraft attached to a mech with drill arms? Works only if:
- mode 1 (where its shorter) is meant to be twice as tall as Mario, and
- mode 2 (standing up) is meant to be three times as tall as Mario (Bowser Jr.'s height included)

Yes, it is as tall as it's meant to be.

Cool! Someday I wanna see a Mario fangame with DK actually in it.
Why? Well no one is gonna make a DK fangame, so he might as well appear with the rest of the Mario cast.

The design, although different from DK, shows a bit of likeness as well. Personally, I love it and it fits his part in the game.
Designed/drawn by Spritanium.
Anyways, I'll probably start on him next. Well... him or the ghost world boss.

Mkay, here we have two concept sprite for the ghost world boss known as "Phangarde"
And for a miniboss that appears later in the game simply known as "Seedy Piranha"
And yeah, this'll probably be the last I'll show in terms of bosses. It wouldn't be any fun if I showed them all now, now would it?

I'd still get C+C, I'd just go about getting it in more private ways, like PMs or on MSN. The main thing is that I'm not the only one working on the game and it wouldn't be fair for me to spoil all of it for everyone else who's working hard in their fields for the game. Anyways, I may post stuff with spoilers from now on.

Seedy Piranha is adorable, but there's a couple weird thing about it that I noticed. Underneath its right arm is some sort of bright glow, like it's holding a flashlight. If that's a spot on its shell, then shouldn't it be covered by the shadow of the hand? Also, its mouth looks taped on. This is just a suggestion, but maybe you should remove the aa and shading between its mouth and face so that it looks more natural.

The "bright glow" is the rim of the shell where it's broken, the flat surface of it would be angled to a point where it would probably be slightly lighter than the rest of the shell. If you look at the other broken points, you'll notice that they all have a bright rim around them, unless I'm misunderstanding what your point. Even if I made that part darker due to the arm, it would still be lighter than the area of shell around it.
As for the mouth, I don't think removing the AA will work. In fact, I tried it and it looked... bad, like his mouth was painted on, but I tried something else and I like it.
EDIT- To help show what mean with the shell, it's meant to be something like this:
Yeah, the arm probably would have more impact on the shading over it, but... idk, I'm hesitant to add extra shading in that small area as I think it may make it harder to read.
