The alpha channel is what lets you see through parts of the image. It's part of the image, and you will know if it's there because you will be able to see the checkerboard pattern of the canvas behind the image. If you need more help on this, there are plenty of tutorials on the basics of image editing and photoshop out there.
Take this image for example. If you can put it into photoshop and export it again with the same transparency, you'll know you're doing it right.
