This scene is the absolute saddest I've seen, not only in gaming but anywhere. Never has any movie or other form of media made my eyes truly well up with tears, but this certainly did. I know that's nerdy, but knowing how gentlemanly the professor is, and how much self control he has, his reactions in this bit are absolutely heart wrenching.
Even now, more than a year after initially watching it, my stomach lurches with the sadness in this scene. I still feel deeply for both Claire and Hershel, despite them being fictional.
I know, I'm way too emotional for a guy.

I can't play any Layton game anymore without feeling sorry for him, this ending is indeed really sad, which makes the game even better. I just wish they would play the ending with lyrics instead of instrumental, it's a beautiful song
I can cry watching anime or reading manga, but no so much while playing games. The only time I can think of getting really emotional because of a game was on Disgaea (probably there are more, but I can't remember them now).
When his mom was the red Prinny, that was actually heartwarming. Also, the whole ending of him wanting to sacrifice himself for Flonne, among other things.
