If you can make it transparent, that will help. If not, definitely pick a color for the background that is not on the sprite.
Second, don't have a lot of empty space. The tighter you can put them together the better. What helps me is TexturePacker. There's a free version and with a little playing around it will create an optimal sprite sheet for you.
