theoretically, yes
though you'd need separate microcontrollers as he did
if I read it right, he didn't repurpose/hack the organ's original music making parts. Instead, he just hooked the keys, switches, jacks, and the speaker up to his microcontrollers. Basically, it isn't so much reverse engineering as it is modding, but whatever.

Oh my God he played Commando's ending theme....That was my second favorite game for the Nes.
